    免疫共沉淀（Co-Immunoprecipitation）是以抗原抗體之間的專一性作用為基礎(chǔ)用于確定兩種蛋白質(zhì)在完整細(xì)胞內(nèi)生理性相互作用的有效方法。當(dāng)細(xì)胞在非變形條件下被裂解時(shí)，完整細(xì)胞內(nèi)存在許多蛋白質(zhì)-蛋白質(zhì)間的相互作用被保留了下來。如果用預(yù)先固化在agarose beads上的蛋白質(zhì)A的抗體免疫沉淀A蛋白，那么與A蛋白在體內(nèi)結(jié)合的蛋白質(zhì)B也能一起沉淀下來，再通過蛋白質(zhì)變性分離，對(duì)B蛋白進(jìn)行檢測(cè)，以證實(shí)兩者存在相互作用。這種方法常用于測(cè)定兩種目標(biāo)蛋白質(zhì)是否在體內(nèi)結(jié)合，也可用于確定一種特定蛋白質(zhì)的新的作用搭檔。
